Welcome to Master of Pharmacy (MPharm). We will study Advanced Pharmaceutics: Preformulation — Solubility, pKa, Partition Coefficient, Polymorphism and Powder Flow, work through an example, and practise using the course's learning material. Solubility and pKa. Intrinsic solubility (S0) is the solubility of the un-ionised form. Most drugs are weak acids or weak bases, so total solubility rises steeply once the pH favours ionisation. The pKa is the pH at which half of the drug is ionised. Example 1 — pH-solubility (invented figures). A weak acid has S0 = 0.1 mg/mL and pKa = 4.0. At pH 6.0, S = 0.1 times (1 + 10^(6.0 − 4.0)) = 0.1 times (1 + 100) = 10.1 mg/mL. At pH 2.0, S = 0.1 times (1 + 10^−2) = 0.1 times 1.01 = 0.101 mg/mL. Check: two pH units above the pKa the drug is 99 per cent ionised, so a roughly hundredfold rise is expected — the answers agree. Which mistake would change the reasoning in this example? Applying the acid equation to a base. For a base, solubility rises as pH falls; the exponent is pKa − pH, not pH − pKa. Treating log P as pH-independent permeability. Ask Course Tutor: please explain this step from the example. At pH 2.0, S = 0.1 times (1 + 10^−2) = 0.1 times 1.01 = 0.101 mg/mL. Why does it follow? Add what you tried, so the explanation addresses your difficulty. Now try the source practice. A weak base has S0 = 0.05 mg/mL and pKa = 8.0. What is its solubility at pH 6.0? Pause before the answer. The source answer is: S = 0.05 times (1 + 10^(8 − 6)) = 0.05 times 101 = 5.05 mg/mL. Compare your reasoning as well as the result. If your response differs, return to the worked example and identify the step that needs correction. Explain the key idea in your own words, revisit any mistaken step, then continue to the next lesson. Use Stop to end Course Tutor when you finish.
